Save states were used in order to achieve this, hence TAS.
Steps:
Spawn Slime at 1600. Spawn Bahamut at 4600. Spawn Slime as soon as the 1st slime hits a peon. Spawn Bullet Train, followed by Fishman and Slime, during the Bahamut hit that occurs after Queen B is KB'd once. Spawn another Fishman ASAP. As soon as Slime is available, spawn Slime, Roe and Green Shell all at once.
Since the strategy here is reliant on Bahamut clipping into Queen B, while simultaneously not having any units go in front of Bahamut, so that Bahamut can get a final hit on Bakoo and do some damage to the base, this strategy isn't practical. However, it was a fun idea, so I gave it a shot.
I'll try figuring out a strategy that isn't reliant on Rich Cat, nor reliant on non-consistent required clipping for a unit.
Levels:
(The first five units are for combos)
Fishman: 40 (lv10 speed talent)
Green Shell: 40
Slime: 45
Bullet Train: 40

This is not a guide. The timings required are too precise for this strategy to be practical. I was just curious if it was theoretically possible to kill puffsley within one bahamut cycle.
Ill try making a more practical strategy though, perhaps with Cat Jobs.
For reference, my unit levels are:
Crystal - 20
Btrain - 50
M. Mohawk - 20
SuperCat - 30
Slime - 50
