Many genetic diseases such as Leber congenital amaurosis can currently not be cured. Gene therapy, however might change that. In the past years, several gene therapies have shown promising results for curing genetic eye diseases. And a few weeks ago, gene therapy was combined with optogenetics to partially restore the sight of a blind patient.

Genetic diseases are caused by mutations in at least one gene. In order to break out, many genetic diseases (but not all) require mutations in the same gene. Leber congenital amaurosis (LCA) is a genetic eye disease where rods and cone cells do not receive photoreceptors from the retinal pigment epithelium (a layer in the retina which nourished surrounding cells). In gene therapy, we try to give the patient an intact version of the mutated gene. To conduct gene therapy, we pack DNA into a vehicle. The vehicle is then injected and taken up by cells. In this manner, the cells receive the intact gene and the disease might be cured.
Gene therapy, however is complex and there are many issues we need to address. Gene therapy should be effective while also being safe. The effects of gene therapy should last for years while not damaging the genetic material of the patient. A few clinical trials have partially improved the sight of LCA patients using gene therapy. The leber congenital amaurosis patients had higher scores in several tests after receiving gene therapy (which led to the production of RPE65). These effects are still not very strong and highly variable.
Optogenetics is a method where we use light to activate neurons. We can give neurons a protein called ChrimsonR which activates the neuron after being exposed to certain light. By combining optogenetics with gene therapy, a study recently successfully improved sight of a genetic eye disease patient. As part of the optogenetics /gene therapy treatment, the patient received ChrimsonR. Then, special goggles scanned the environment and activated ChrimsonR leading to improvements in the eye sight of the patient. This is an indication that gene therapy and optogenetics might be feasible to cure genetic eye diseases.

Today we will talk about 3 common human parasites: these human parasites are Toxoplasma gondii, Giardia & Hookworms. These human parasites have infected numerous people worldwide and therefore many scientists aim to find out how we can prevent infections with these human parasites. By the end of this video, I hope that you remember the three human parasites we will discuss and how they are transmitted.

Lets go trough the human parasites one by one. Toxoplasma gondii is a human parasite which has infected over 2 billion people worldwide. This means that this human parasite can be found on average in each family. The human parasite Toxoplasma gondii is mostly transmitted through cat feces. Once inactive toxoplasma gondii egg-like structures have entered the body, they undergo a transformation a produce many toxoplasma offsprings. After the immune system has recognised toxoplasma gondii, the parasite starts to hide in muscles, eyes and the brain. Toxoplasma gondii can stay in the brain for years. Normally, toxoplasma infections are not detrimental . HIV and cancer patients and unborn babies can develop toxoplasmosis however which is a serious human parasite infection. This is especially dramatic if a pregnant woman becomes infected with toxoplasma gondii. The resulting toxoplasmosis might result in stillbirth or lead to developmental issues. Therefore, Toxoplasma gondii can be a detrimental human parasite and is one of the most common human parasites.
In contrast to toxoplasma gondii, the human parasite giardia lamblia can be found in food and water. Over 220 million people are infected with giardia. Giardia can cause giardiasis which is characterised by abdominal pain and flatulence. Pregnant women and children are at highest risk to develop giardiasis.We can use metronidazole to treat giardiasis.
Hookworms are human parasites currently live inside almost 500 million people. These human parasites are normally contracted by human feces. Hookworms can survive in soil for a long time and infect people who pass by. People who are infected with hookworms see rashes and then experience abdominal pain and diarrhea. Currently, hookworms are studies as they might help to treat autoinflammatory diseases.
