Optimization: tackle jump to the right off the awning(?) over Tuskle's door, then double jump around to land on the back side of the roof. This gets you into the right position for the first Vine slightly faster.
This video demonstrates an alternative method of entering Wawku Shrine early in NG, compared to the existing method (seen here youtu.be/rg3d8MtylbQ?t=8656).
This method abuses the fact that Vine will snap on their own if Ammy is blocked from reaching them for too long. In this case, we use an invisible wall to block Ammy twice, enabling us to get enough height to clear the invisible wall and get behind the gate to Wawku.
This method is pretty comparable in speed to the old method. I doubt either is more than 2 seconds faster than the other. Of the two, this may be more reliable due to the lack of Vine cutting, which can be inconsistent at some angles. Then again, if you start from the wrong position, you might slip past the invisible wall and end up on the flower if you don't react fast enough, which loses 1-2 minutes. This can happen with either method, though, so I'm not sure yet which will win out in the end.
I should note that there's a few different things that can happen at the top when you do the double jump after the second vine. It's possible to get a wall-jump here, but I've only gotten it a couple times. Nevertheless, that makes this hypothetically possible in a no-double-jump run. You might clip over the cliff entirely if you're not careful, which can result in falling into the void (or worse: falling onto the OoB ice). You may even become grounded long enough to get a ground tackle off, though this isn't useful.

My first PB in over 2 years! Ive had very little time to run in all this time, but I was finally able to practice enough to not flub the huge advantage I had over my last PB from all the new tricks and optimizations discovered since then.
Theres a lot to improve from here. In terms of major time losses, those are:
-Ryo Stair Clip (30s)
-Blockhead Clip (2m)
-Fire Tablet Skip (3m)
-Shinshu Plateau Demon Boost (47s) (I actually got this really quickly... and then slipped off the plateau trying to jump up to the thunderdome )
-Oki Skip (I lost 70s on this split, though a big part of that was trying to figure out a way to lose negative godhood)
-Wawku Cannon Room (1m)
A lot of the rest of the time loss comes down to general sloppiness: suboptimal movement, early/late cutscene skips, bad fights, slow brushing, etc.. That level of confidence will come if and when I do more and more runs.
Turbo start was used to skip cutscenes. Turbo jump was used primarily to swim faster and occasionally to jump immediately after landing.
