The standard strat for defeating the Spider Queen in Tsuta Ruins is to immediately Vine her abdomen twice, wait until the flowers disappear, then immediately use 2 Small Slips (ExS) followed by 3 Power Slashes through all the eyes on her back.

This video shows an alternate method, where we only use 1 ExS. First, Vine her once, then wait until your 5th ink pot refills (you should always have 5 by now, in both NG Any% and All Brushes), then Vine again. Now, PS her eyes 4 times, instead of the usual 3.

If you miss any of the eyes in those 4 PS's, you'll have time to run up to her abdomen and manually attack them, though if you're too inaccurate you may get swallowed. This method is fairly lenient on timing, at least.

Compared to using 2 ExS, this strat takes about 6-7s longer if both are done perfectly. Considering it takes about 6s to pick up either of the 2 fastest ExS, this won't save any time in NG Any% (at least not more than a second or so), but it should be worthwhile in NG All Brushes, where we can find a better use for that second ExS against Crimson Helm.
