The Castle already existed at the beginning of the 13th century, with a layout very similar to the current one. In the early Middle Ages, the property of Trisobbio was divided in three between the Marquises Del Bosco, those De Uxecio, that is of Belforte, and those of Occimiano. In 1217 the Marquis Ottone Del Bosco and his nephews donated Trisobbio to the municipality of Genoa. The situation of co-ownership between Alessandria, which had received marquisate rights from the Marquises of Occimiano, and Genoa resulted in a war in 1224 during which Trisobbio was occupied by the Alessandrian militias who settled in the Castle. In the subsequent peace negotiations it was decided that the fortification had to be returned to the Del Bosco family and therefore, indirectly, to Genoa. In 1240 Frederick II, looking for allies, validated the ancient rights of the Marquises of Occimiano and shortly afterwards invested the Marquis of Monferrato as feudal lord of Trisobbio. The town thus continued to be divided between various feudal families. The issue was resolved when the Malaspina, who had succeeded the Del Boscos by inheritance, received recognition from both the Marquises of Monferrato and Genoa. During this period, the Castle and the village underwent further modifications, necessary to improve its defense. In 1418-19 Trisobbio was occupied by the Milanese, during the war with Genoa and by them handed it over to Monferrato, their ally. The fiefdom then passed to another Genoese family, the Spinola, who held it until the Napoleonic invasion. Around the end of the 19th century, the structure was in a state of degradation and ruin. In 1913, the Marquis Carpaneto Spinola asked, through the Superintendence of Monuments of Liguria, the architect Commendator Alfredo D'Andrade for the documents to begin the restoration work. This work brought the Castle to its current external appearance.
