Stage 5 is a very unique one, and plays heavily into the “risk vs reward” aspect of this game. Effectively you've got these “trap zones” around the field that'll explode with bullets if you touch them. They're easy enough to avoid so long as you leave yourself plenty of room to get away after you trigger them, however if you actually manage to avoid them all completely, you'll be rewarded with life pieces depending on how many are still standing when Yamame returns each time. Yamame herself isn't much to write home about though, since she's ultimately pretty easy once you understand what you're getting yourself into. All her attacks are essentially aimed at you, so that can lead to you accidentally trapping yourself, but if you understand the logic of it, you can keep moving to manipulate the spell to always leave yourself an opening.
Kisume's not too bad, once you know what you're getting into anyways. She's one of those characters who's not so much difficult, but rather she's tricky and can catch you off guard with surprise attacks, or a spell not quite acting like how you expected it to. Learn her spells and what she does however, and she becomes much easier to deal with.
