This is a Tool-Assisted Superplay/Speedrun (often abbreviated to "TAS") where a player can edit, rerecord, and manipulate a game into doing whatever that person desires. Visit tasvideos.org for more information.
This is an improvement to my 1.88 from a few years ago. Optimization for this stage challenge is to simply have 2 golden Chu Chus enter your rockets as fast as possible, but even just one is very rare. As a method for manipulating RNG, I made a stage in the stage builder that has Chu Chus constantly running into rockets. Interestingly, because RNG doesn't advance when not called, the Chu Chus that would have come out in the stage builder could instead come out of the rockets in the stage challenge if I quit beforehand. Essentially, I would wait until I had two golden Chu Chus come out in close succession, and then hope they would also come out of the right side rockets in the stage challenge.
After about half a million frames worth of RNG calls through this method, this was the fastest that I found. A bit more time could hypothetically come off, but I couldn't find such RNG to make that possible.

The precision for this is inhuman, and barring a new discovery or me having overlooked something, this will almost certainly never be achieved for real. This is basically the same as my setup TAS, except I do a very precise adjustment to get 10 extra points. I didnt want this to be the example video for my setup TAS because high scores were a lot less dense. Of all combinations of X and Y adjustments of the controller stick, this was the only one that I found that gave this score. After many hours of research due to how rare the required RNG is and how precise the shot is, I was able to create this setup to allow for higher scores than the older style of shots. Be warned though that this strategy is extremely difficult and I would not recommend it for most players.
Instructions:
1) Reset the console, select Tunnel, skip the cutscene.
2) Turn around facing backwards and throw 17 pester balls, let the gas of the last one fade out, then pause and select retry. Speed isnt an issue as long as you arent entering the cave or doing anything weird.
3) Skip the cutscene, hold turbo, crash into Electrode, turn to the right, and continue holding turbo.
4) Pause on the correct frame, release R, and hold Z. Pausing on this frame is mandatory or the strategy wont work.
5) Aim a bit upwards, then press C-left when youre ready. You can use the slopes of the wall as a visual cue and you can turn at any time.
6) Start aiming a bit to the right and press A on the correct frame to make the shot.
Tips:
-Because Kakuna falls so quickly and the game animates at 60 frames per second while running at 30 frames per second, half of all attempts will be impossible to get a good shot of Kakuna. You must simply overcome this by grinding out attempts.
-For knowing youre on the correct pause frame, look at the line between the blue and brown spots under the retry button and make sure its at the right place. Dont waste your time practicing the wrong pause frames.
-For adjusting horizontally, theres a particular dark spot on the wall that you can aim for.
