*Update 1*: Can confirm this even works on NG+5. Used a x5 Multiplier and nuked Simon in Phase 2. Of course on the NG+5, you will have to manage the speed bit with Greater Slow on Simon.
You guys were asking if any of the Endless Tower Super Bosses can be one-shot? Well, here is Simon the Divergent Star. This is on NG but I used the x2 Health Multiplier which you can see at the start of the battle to show that this strategy can be done even on NG+ 1 and 2.
Used some attacks to build up the needed buffs, and then one-shotted Simon out of his Divergent Star. You may say that this is technically not a one-shot, since I used other attacks. But at the end of the day, he got nuked in both phases.
