The answer is two. The game requires the drummer or it'll crash. Singer and bassist are totally optional.
The idea for this one partially came from my girlfriend posting Welsh harp music that was like ten minutes of crazy soloing and my sudden desire to chart something like it. I don't have any of that in lossless right now though, so I chose this roughly minute-long guitar instrumental from the Oddity Archive Audio Library. You don't even need a bass part, just copy the guitar part and make it a rhythm part and two players can just play the same piece in co-op. That way, all modes work with a solo instrumental.
This VGS also only has three channels in it, the stereo guitar and then a mono band. Mapped correctly, I think you can just have a VGS with a single channel shared by both players with no band stuff to speak of (as "band" isn't mapped, the game just plays all unmapped tracks as background noise). So really, this whole thing was an experiment in just how minimal a custom the game will accept.
The song is of course "Souls for Sale", from Ben Minnotte's (remember my "Dear Kate" custom?) "Greetings From Elderbush Gulch", which you should take a listen to and potentially buy here if you dig it: benminnotte.bandcamp.com/album/greetings-from-elderbush-gulch-2011-remix-remaster
And of course, like I said the other day, that'll do it for the single chart releases! My eyes are now set on chart sampler discs. If you want some of those (this chart is on marfGH: Volume 0), go check them out at my website: mariteaux.somnolescent.net/modding/guitar-hero/discs

Figured a good way to get back into the marfGH swing would be with a classic (to me).
Watch and listen closely! Theres actually two minor hacks I did up for this (audience clapping if youre doing really well and pre-chart end cheering like [music_end] in Rock Band), plus showing off more of my custom camera cutting functionality. It takes a lot of tweaking, and maybe Im giving myself a lot of extra unnecessary work for GH2, but its worth it.
Let me know if that strumming is overcharted, I dont think it is.
