After officially unlocking the empire sites by taking over Marty's businesses, you can start missions by heading inside a business or upgrade it for higher security and pay rate. Currently, Protection Racket, Loan Shark and Pimping will be possible to complete. After unlocking the second Island, along with taking over an enemy business that is Robbery, Drugs or Smuggling, you'll unlock those.
Remember to build one of each business types at any site up to High Roller, since that'll also count toward 100% completion.
