I found a TAS of this and decided to improve it a little bit. If you have Diddy in the start you'll be able to finish Very Gnawty and Queen B faster than with Donkey. He is needed against Dumb Drum and is faster against Really Gnawty. After some testings, starting with Donkey was faster overall.
To finish Queen B the fastest she has to be as low as possible (Too low and she'll fall through the floor though, which is slower). To manipulate this I had to delay one of the hits by one frame.
Committing suicide against Really Gnawty skips his last high jumps so you can hit it directly. To die against the drumb is also intentional, as I need Donkey for the last two rounds. If I then break the DK barrel against the final two bosses, I'll lose time. To avoid breaking it you can use a groundslap with Donkey and then jump.

2-1: While there is an easily accessable Bonus Room, you arent actually shot out from a barrel after the bonus, so there is no speed you can keep throughout the level.
2-2: Pretty much the same as regularly.
2-3: Entering the Winky room to build up the speed. Winky does not move any faster, he would only cost time to get.
2-4: I like how I didnt have to lose any speed to avoid the Rockrocs, worked out nicely!
2-5: You can only reach the Warp Barrel with Donkey and setting this up was a bit tricky to do fast as I need to keep jumping to keep the speed and pushing the tire at the same time was dumb.
