Digital PCR (dPCR) can provide absolute quantification, without standard curves, along with unrivaled accuracy and precision. With more dPCR providers than every in the marketplace these is a lot of talk about which dPCR system has the lowest dead volume, which matters when there is limited sample volume and/or low copy number target in the sample.
Dead volume is the portion of the sample that’s not directly used in the analytical analysis. Volume can be lost in collection, preparation, and in the dPCR analysis process itself. Essentially, the entire dPCR workflow needs to be considered when discussing dead volume.
The early dPCR workflow steps of sample collection and sample preparation are often neglected but should not be as sample volume and target template copy number can easily be lost here. Optimization of these steps should be considered, regardless of dPCR platform being used.
In the dPCR portion of the workflow every provider partitions the sample, amplifies the targets, and then reads and analyzes the sample. However, Bio-Rad™ Droplet Digital™ PCR (ddPCR™) does things differently. Specifically, only ddPCR technology uses emulsion-based partitioning and only ddPCR technology offers gradient capabilities in the PCR amplification step. Droplet partitioning creates up to 20,000 sample partitions, regardless of consumable choices, to ensure that under sampling errors are avoided. The gradient capabilities of ddPCR technology allow for faster PCR optimization to minimize sample used during optimizations steps, a consideration often overlooked by others.
Digital PCR dead volume is important in situations with limited sample and applications with rare targets. For these situations consider the entire dPCR workflow to optimize early steps and select a dPCR platform that reliably makes the most of your precious samples.
