The synthesized “awakening” motif appears at the start of “316” as a direct reference to the show's opening scene, aiding the visual similarities between both moments. It's also used as a musical ‘bookend’ for the season 4 finale, which, again, involves Jack Shephard opening his eyes when waking up, as well as a suggestion to return to the island.
Unlike the rest of the show's score, the “awakening” motif is evidently inspired by original temp score (specifically Vangelis’ “Blade Runner” soundtrack) used for the Pilot episode. Future episodes of LOST would feature temp tracks composed by Michael Giacchino himself, but he would never listen to it, believing it would limit his creative process.
