A solution of a soluble protein is treated with concentrated nitric acid. The formation of a yellow color is the prove of the existence in the solution of a protein. The aromatic rings of Phe and Tyr are nitrated giving birth to a yellow complex which, upon alkalinisation, turn to orange.
