1. In the Sanctuary of Rock and Lava, there's a room with a spinning platform and a lavafall. After climbing the wall and landing on a stationary ledge, jump on the ledge that moves around the spinning platform. Then get hit by the lava, go through it, and grab a nearby edge. This saves a few seconds over the normal route.
2. In the Sanctuary of Stone and Fire, there's a room with a squeaky pirate and a plum. With a well-aimed throw, you can take the plum to the previous area, which seems like it would save time in a 100% run. However, the plum is actually much slower than the plum you would normally use in that area, so using the normal plum would probably be faster.
