As you can see, I'm a little shaky here; but I never lose a life and still get the colored gem in the end.

In the first couple of seconds in the video, you can probably tell that I lost a life before starting this Perfect Run. From this playthrough I found out that you are allowed to lose lives before you reach the first checkpoint and still qualify for the gem.
