The goal is to reach the end of the game as soon as possible. For the most part, this means playing through the game quickly and avoiding all optional things, such as talking to Oskar more than once.
The major skip I discovered happens at 33:41. Before the second death scene, I save the game. I then load a save file, at the end of the game. I trigger the final autosave of the game. I go back to the saved game, and I purposely die on the second death scene. Instead of sending me back to the start of the death scene, the game sends me to the autosave. This basically skips the final hour of the game!
At 9:58, I look at the artifact and run out of the building, before Oskar can talk to Nancy about the artifact. Not only does that skip a conversation, but it also skips the Oskar conversation at 23:23, which normally occurs when you first open the artifact.
I missed a skip at 20:49. In the brief second after Cyber Block ends, you can click on Elka, then go to the main menu and resume. This tricks the game into thinking you're having a normal conversation with Elka, so it skips over the Zane conversation.
At 33:08, I skip the alchemy puzzle by entering the correct safe combination.
The speedrun can easily be improved, by playing the game on Classic Mode. Classic Mode is faster, when going through the big courtyards. I didn't play it on Classic Mode, because I didn't want to spend a lot of time memorizing the navigation.

Master Sleuth mode does not include a task list or the hints that appear on the task list. Five puzzles have differences; they are cyber block, the candles puzzle, the safe puzzle, the text riddle puzzle and the bottom part of the clock puzzle.
With the safe and text riddle, the difference is with the clues, as the solution is the same on both modes. Coffee making tends to be more difficult on Master Sleuth mode, as you are more likely to get harder orders.

Two characters appear, in time to see the culprit's arrest.
The day after, Agent Zane Garrett appears in person! He helps wrap up the various loose ends involving Rezidente and Loutkaji. Nancy's phone and Adela's necklace are returned. The game comes to an end, when Nancy calls either Ned or her father. (I included both endings in this video.) Mystery solved!

They're going to follow the culprit's orders by leaving the keys near a statue, but with a twist: one of the keys has a tracker inside!

When leaving, Nancy has another encounter with the shadowy knight. She leaves the journal in the cafe, so Adela can translate it. She talks to several people about the Rezidente, including Ned and Oskar. She calls the police for an update on their search for Oskar. She wraps things up, by talking to Radek. That brings an end to her investigation for the night.

The mayor says the only way to get into the castle is to collect two stone keys from Shhwonk Fortress. The mayor gives permission for Mario to go there. Just as Mario is about to leave, a koopa named Koops tries talking to him. Our heroes go to the ruined city underneath Rogueport, where they find some minor enemies and the Thousand Year Door. Mario finds a key that unlocks a chest, only to be cursed with the ability to turn into a paper airplane. Luckily, this curse is more useful than harmful.
