There are several things to take particular note of. First, changes to the pressure applied to the pipette tip can be seen as changes in the dye streaming out of the respective tip. Next, dimple formation can be observed in both images for each pipette, but it is most pronounced for the third cell. Talk Abstract Commonly, fluorescence microscopy uses the colour and intensity of fluorescence to generate images. A third parameter of fluorescence is less commonly used in imaging: the excited state lifetime of a fluorescent molecule (aka the fluorescence lifetime). Similar to colour, the fluorescence lifetime is an absolute value which is not influenced by properties of the measurement system or by local concentrations of the fluorescent dye. As a result, fluorescence lifetime imaging (FLIM) can provide quantitative insights into the physiology of live cells. In combination with two-photon excitation, it becomes a powerful tool for the quantitative study of cellular signalling in deep tissue.
