At AGBT 2026, Dr Bodour Salhia PhD (USC Norris Comprehensive Cancer Center) presents the development and clinical validation of OvaPrint a cell-free DNA (cfDNA) methylation–based liquid biopsy designed to improve ovarian cancer risk assessment in patients with adnexal masses.
Current diagnostic approaches rely on protein biomarkers with limited specificity, often leading to unnecessary surgical procedures. OvaPrint addresses this gap, achieving 96% specificity and strong sensitivity for high-grade serous ovarian cancer, the most common and lethal subtype.
This talk highlights how Illumina 5-base chemistry enhances methylation signal resolution in plasma, improving detection of non–high-grade serous ovarian cancers that are traditionally harder to classify. To further strengthen performance, the study integrates multiomic approaches, including plasma proteomics using Illumina SomaSeq Discovery and spatial transcriptomics to provide tissue-level context.
Results demonstrate that combining methylation, proteomic, and spatial transcriptomic data improves interpretation of challenging cases, reduces false positives and negatives, and provides a more complete biological understanding. This multi-modal framework represents a promising strategy to improve clinical decision-making and reduce unnecessary interventions in ovarian cancer care.
