Description:
The ATP Synthase motor's job is to manufacture a little molecule called ATP - short for Adenosine triphosphate - which is of enormous importance for the successful functioning of our bodies.
ATP synthase (EC 3.6.3.14) is an important enzyme that provides energy for the cell to use through the synthesis of adenosine triphosphate (ATP). ATP is the most commonly used "energy currency" of cells from most organisms. It is formed from adenosine diphosphate (ADP) and inorganic phosphate (Pi), and needs energy.

The overall reaction sequence is: ADP + Pi → ATP, where ADP and Pi are joined together by ATP synthase

Energy is often released in the form of hydrogen ions (H+
), moving down an electrochemical gradient, such as from the lumen into the stroma of chloroplasts or from the inter-membrane space into the matrix in mitochondria.
Like any other motor it rotates, and surprisingly fast - in fact at about 6,000 RPM (revolutions per minute) ! Further, it is the last word in ultra-miniaturisation, being 200,000 times smaller than a pinhead! And we have rather more of these motors than most vehicles! Indeed, every cell in the body has hundreds, if not thousands of them. As we have some 100 trillion (1 followed by 14 zeros) cells, there are in excess of 10 quadrillion (1 followed by 16 zeros) of these amazing ultra-tiny little motors which drive our bodies and upon which our very lives depend!

Talk overview: Berg begins his lecture with a brief history of observations of bacterial motion. He then uses physics to describe the many hurdles that E. coli must overcome as it tries to swim up or down a chemical gradient. For instance, an entity as tiny as E. coli is constantly buffeted by Brownian motion and can neither stay still nor swim in a straight line. Then there is the question of how E. coli senses a gradient and translates that information into a change in its direction of movement. And finally, how does E. coli use its flagella to generate thrust at all?

In Part 2, Berg explains that E. coli travels using a series of runs, when it moves in a straight line, and tumbles, when it changes direction. During a run, all of the flagella are moving counterclockwise in a tight bundle. During a tumble, one or more flagella switch to a clockwise movement and disengage from the bundle causing a change in the swimming direction. The motor that drives the rotation of the flagella is an amazing structure made of about 20 different protein parts. Berg tells us that chemosensory receptors on the cell surface detect a chemical gradient and transfer this information, via protein phosphorylation, to the motor. This chemical modification determines the direction of motor rotation and, hence, the direction the E. coli swims. An amazing system that E. coli has been perfecting for millions of years!

Speaker biography: Howard Berg is the Herchel Smith Professor of Physics and a Professor of Molecular and Cellular Biology at Harvard University and a member of the Rowland Institute for Science at Harvard. He received his B.S. in Chemistry from the California Institute of Technology and his Ph.D. in Chemical Physics from Harvard University. Berg was on the faculty of the University of Colorado and Cal Tech before joining Harvard in 1986.
Berg's lab applies methods from physics to biological problems. They strive to understand how a bacterium, such as E. coli, can sense changes in its environment and respond by swimming towards, or away from, a stimulus. To this end, the lab studies the mechanics of the bacterial flagellar motor and how it is regulated by signals from cell surface receptors.
Berg has received numerous awards and honors for his work including election to the National Academy of Sciences and the American Academy of Arts and Sciences.

What is something computers and humans have in common which constantly needs upgrading in computers but not in humans? The answer is software. You might not have realized that you have software but inside the nucleus of each of your cells a program is written in the form of three billion DNA letters.

Intelligent programmers write computer software but what about living things? Evolutionists tell us that the information in the first living cell just appeared by itself, with no intelligent input required. But is that possible? The answer is a resounding no.

Even one of Australia's best known scientists, Paul Davies, conceded that, "There is no know law of physics able to crate information from nothing." And perhaps that's why, in a New Scientist article, he lamented, "How did stupid atoms spontaneously write their own software? Nobody knows."

Cells harvest usable energy from food using organelles called mitochondria. Inside the mitochondrion, there is a highly folded membrane that contains the electron transport chain. As high energy electrons from food are passed down the chain, protons are pumped out of the membrane. These protons flow back through a molecular machine called ATP synthase to make ATP and power the cell. Cyanide is a very deadly poison because it interferes with this process, which is essential for human life.

ATP, or Adenosine Tri-Phosphate, is the energy currency of the cell. ATP is produced by a tiny molecular rotary motor, rotating at up to 7,000 rpm. These are so small that 100,000 would fit side-by-side in a millimetre. A current of protons drives the motor, unlike man-made electric motors, which use electrons.

This portion of the enzyme is where Adenosine Di-Phosphate is combined with a phosphate ion, in the presence of a catalyst to produce ATP which is then released, making way for the next cycle. A top view of the enzyme shows the sequential operation. Almost every bio-chemical process in your body requires ATP.

Such a nano-machine exhibits all the characteristics of super-intelligent design. ATP is vital for life and many of these motors were needed before the first living cell could exist. An evolutionary impossibility!

BEHE CONCEDES IC SYSTEMS CAN EVOLVE

Here I present a theory first published in 2003 by Nicholas Matzke. For more details one can view his work at

This theory is derived from a wealth of data regarding protein homologies. It shows that a fully functional flagellum could evolve through multiple functional intermediates. Each step involves the modification of only a single protein. For more discussion on the actual proteins involved and their homologues see the link above.

When ID proponents ask what good is half a flagellum, well, ¾ a flagellum is good for dispersion, ½ a flagellum is good for attachment to substrates, ¼ a flagellum is good for regulated protein secretion.

This model based on a wealth of evidence shows that the flagellum could evolve. Since 2003 many experiments have confirmed what initially were only predictions or weak assumptions. Still, many fine details remain to be worked out regarding function, not origin. For example, how does the ATP synthase derived components impart the motion of active transport, or how does the Tol-Pal derived components actually spine the flagellum.

Behe in the Dover trial concedes that Irreducibly Complex systems can evolve.

Lawyer -- "You say, Even if a system is irreducibly complex
and thus could not have been produced directly, however,
one cannot definitively rule out the possibility of an
indirect, circuitous route, right?
Behe -- "Yes."
Lawyer -- "And by indirect, you mean evolution from a
pre-cursor with a different function than the system
being studied?"
Behe -- "Yes, different function, perhaps different number
of parts, and so on."

To be fair, however, Behe claims that at a certain point of complexity an indirect route is impossible. But he gives no evidence to support this caveat.

The animation highlights the creation of Adenosine Triphosphate (ATP) -- mobile molecules which store chemical energy derived from the breakdown of carbon-based food. ATP molecules act as a kind of currency, imparting chemical energy to power all the functional components of cellular activity. This piece is the second in a series of award winning animations XVIVO is creating for Harvard's educational website BioVisions at Harvard. The first program, Inner Life of the Cell, received international acclaim and can be seen both on our website and the BioVisions site.
